Lee Yu Ri mulai dikenal naanya melalui sebuah drama berjudul School 4 yang ditayangkan pada tahun 2001. Walaupun begitu, ia telah mulai mencoba berkarir di dunia akting pada tahun 1990-an.

Kemudian pada tahun 2005, namanya semakin banyak diperbincangkan karena berhasil membintangi drama Precious Family. Pada tahun 2011, ia kembali dengan drama Twinkle Twinkle.

Setelah itu, ia tampil di berbagai drama lainnya, seperti Spring Turns to Spring yang tayang pada tahun 2019. Dilanjutkan dengan drama lainnya berjudul Lie After Lie (2020).

Sukses membintangi berbagai drama tersebut, akhirnya ia kembali dengan karakter utama untuk drama berjudul Becoming Witch (2022).
