Amanda Seyfried memulai karirnya di dunia hiburan dengan menjadi model iklan. Saat itu, ia menjadi model untuk perusahaan pakaian Limited Too dengan Leighton Meester. Bahkan ia juga pernah tampil dalam sampul novel Sweet Valley High.

Namun, ia kemudian berhenti menjadi model saat berusia 17 tahun dan mengambil pelajaran vokal. Selain itu, ia juga belajar opera serta berlatih broadway saat masih remaja.

Ia pun akhirnya debut akting di tahun 1999 dalam sebuah serial TV berjudul The World Turns (1999-2001).

Dalam serial tersebut ia berperan menjadi Lucinda “Lucy” Montgomery. Ia juga pernah tampil dalam 3 episode di serial All My Children (2003).

Namanya menjadi terkenal setelah ia tampil sebagai Regina George dalam film berjudul Mean Girls (2004).

Namun pada akhirnya, ia berperan sebagai Karen Smith dalam film Lindsay Lohan tersebut.

Setelah itu, ia banyak membintangi film lain berjudul American Gun (2005), Mamma Mia! (2008), Letters to Juliet (2010), A Million Ways to Die in the West (2014), Scoob! (2020) dan banyak film lainnya.

Selain itu, ia juga membintangi beberapa judul serial TV diantaranya Law & Order: Special Victims Unit (2004), CSI: Crime Scene Investigation (2006), Twin Peaks (2017) dan banyak lagi.
